    A.1.1 :水

    1.1

    既知の値から始める

    • 既知の値と単位を明記する。このケースに適応する仮定を持つ関係式を選び、代入前に変形する。
    • 既知:0.100 kgの水が5.0 K温まる;比熱容量は4,180 J/(kg K)。Q = mcΔT = 0.100 × 4,180 × 5.0 = 2,090 J。この計算は容器が吸収したエネルギーや周囲へ失われたエネルギーを含めない。

    例:

    c = 4,180 J/(kg K)を使用する。0.200 kgの水を5.0 K加熱した場合のQを計算する。同じ手順を用いる:既定量 → モデル → 関係式 → 代入 → 単位と解釈。

    結論とその限界を確認する

    • 分子間の水素結合は、水分子内の共有結合とは異なります。極性だけでは、すべての物質が水によく溶解するわけではありません。
    • 元の観察に戻り、結果が何を裏付けているか、どのような条件を前提としているかを説明し、対立する説明を検証する方法を一つ示す。

    警告:

    沸騰すると水は通常O–H共有結合を切断する。この主張は誤りである:分子間の水素結合は、水分子内の共有結合とは異なる。極性だけでは、すべての物質が水によく溶解するわけではない。

    重要:

    水:生物学的性質のための分子的要因:同じ分子間相互作用は表面張力や温度変化に必要なエネルギーを説明するのに役立つ。加熱は分子運動と相互作用を変化させるが、通常O–H共有結合を切断しない。

    2.2

    DNA、タンパク質合成およびその根拠

    この観察を説明するのは何か?

    • DNAの変化はタンパク質に影響を与えることがありますが、すべてのDNA変化がアミノ酸配列を変えるわけではありません。その効果は配列とそれがどのように利用されるかによって異なります。
    • まず予測を立てる。比較したい量や特徴を明記し、2つの説明を区別できる証拠は何かを決定せよ。

    モデルを構築する

    • DNAは塩基配列で情報を保持しています。相補的塩基対結合は複製を支援します。遺伝子発現中、転写によりRNAが作られ、翻訳ではコドンを用いてアミノ酸配列が組み立てられます。
    • 転写:DNAテンプレートを用いたRNAの生成;翻訳:mRNA配列を用いたポリペプチドの生成。
DNA, protein synthesis and evidence: original worked-case diagram

    Scope and prerequisites

    English

Supported SL core focus. First assessment 2025; current subject brief acquired; full Biology guide not acquired. Remaining guide, assessment and practical requirements retain their recorded holds.

Prerequisites: amino-acid structure and condensation reactions from B.1.1 (carbohydrates and lipids), and microscopy units from B.1. Each lesson develops its own method before independent transfer.

These are original or explicitly fictional teaching examples, not actual measurements or completed assessed learner investigations.

    8.2

    Proteins

    English

    What would explain this observation?

    • An egg white turns from clear runny liquid to firm white solid when heated — and cooling never turns it back. What happened to its proteins?
    • Start with a prediction. State the quantities or features you would compare, then decide what evidence could distinguish two explanations.

    Build the model

    • Proteins are chains of amino acids joined by peptide bonds 肽键 in condensation reactions; each bond forms with the loss of one water molecule. The chain order is the primary structure 一级结构, which folds into local helices and sheets (secondary), then into a working three-dimensional shape (tertiary), sometimes with several chains together (quaternary).
    • The twenty R groups differ in charge and polarity, so the sequence decides the fold. Heating or extreme pH breaks the weak bonds holding the fold — denaturation 变性 — while the peptide bonds survive; the protein unravels and loses its function, which is why the egg does not run again on cooling.
    • peptide bond: The covalent link between two amino acids, formed with the loss of water; primary structure: The sequence of amino acids in a protein chain; denaturation: The loss of a protein's working shape when its weak structure bonds break.
    Proteins: original worked-case diagram

Choose evidence that can test it

    • The Biuret test identifies peptide bonds: it turns from blue to purple with protein but not with free amino acids. Use it to check whether a sample contains peptide linkages.
    • Compare an enzyme's activity before and after heating, keeping substrate and pH constant: a lasting loss of activity after cooling supports permanent denaturation, not just a reversible pause.

    Work from known quantities

    • State the known values and their units. Choose the relation because its assumptions fit this case, then rearrange before substitution.
    • Known: a peptide chain contains 35 amino-acid residues joined in one unbranched chain. Each link between two neighbours is one peptide bond, so bonds = residues − 1 = 35 − 1 = 34. Each bond formed by condensation releases one water molecule, so 34 water molecules were released when this chain formed.

    Example:

A different chain contains 120 amino-acid residues. Find the number of peptide bonds and the number of water molecules released during its formation, using the same sequence: known quantities → model → relation → substitution → unit and interpretation. (Answer: bonds = 120 − 1 = 119; waters released = 119. Hydrolysis back to single amino acids would need 119 water molecules.)

Check the conclusion and its limits

    • The bond count assumes a single unbranched chain; real proteins may be branched or cross-linked by disulfide bridges, which are not peptide bonds. Denaturation is usually irreversible in food but some small proteins can refold under careful laboratory conditions.
    • Return to the original observation. Explain what the result supports, which conditions it assumes, and one way to test a competing explanation.

    Warn:

Heating denatures a protein by breaking its peptide bonds. This claim is false: denaturation breaks the weaker hydrogen bonds and interactions that hold the fold; the peptide bonds of the primary structure survive, which is why the amino-acid sequence is unchanged in a cooked egg.

    Key:

Proteins: amino acids join by peptide bonds through condensation, releasing one water per bond (n residues → n−1 bonds). The sequence folds through secondary and tertiary structure into the working shape; denaturation destroys the fold, not the chain.

    Scope and prerequisites

    English

Supported SL focus. First assessment 2025; current subject brief acquired; full Biology guide not acquired. Remaining guide, assessment and practical requirements retain their recorded holds.

Prerequisites: read the stated quantities and units, use arithmetic and the model conditions below. Each lesson develops its own method before independent transfer.

These are original or explicitly fictional teaching examples, not actual measurements or completed assessed learner investigations.

    9.2

Osmosis 渗透作用 and a fair potato experiment

    English

    What would explain this observation?

    • A potato cylinder gains mass in one solution and loses mass in another. Its mass change provides evidence about movement of water, rather than movement of potato tissue.
    • Start with a prediction. State the quantities or features you would compare, then decide what evidence could distinguish two explanations.

    Build the model

    • Osmosis is the net movement of water through a partially permeable membrane from a more dilute solution to a more concentrated solution. Dissolved solutes can change the direction of net water movement.
    • osmosis: Net water movement through a partially permeable membrane; control variable A factor kept constant for a fair comparison.
Osmosis and a fair potato experiment: original worked-case diagram

Choose evidence that can test it

    • Use percentage change to compare samples with different initial masses. A zero percentage change estimates a solution concentration with no net water movement. This is an estimate from a trend, not proof that water molecules stop moving.
    • Use equal-length cylinders from similar tissue, fixed solution volume, temperature and immersion time. Blot each cylinder in the same way before weighing. Repeat each concentration and plot mean percentage change against concentration.

    Work from known quantities

    • State the known values and their units. Choose the relation because its assumptions fit this case, then rearrange before substitution.
    • Known: initial mass 2.50 g; final mass 2.75 g. Use percentage change = (final - initial) / initial × 100. Percentage change = (2.75 - 2.50) / 2.50 × 100 = +10%. A positive value means net water entry. Interpolate the concentration where the plotted trend crosses zero.

    Example:

A 4.00 g cylinder ends at 3.60 g. Find percentage mass change. Use the same sequence: known quantities → model → relation → substitution → unit and interpretation.

Check the conclusion and its limits

    • Blotting removes surface solution. Weighing a wet cylinder without blotting adds liquid that did not enter the cells. Repeats reduce random variation but do not correct a miscalibrated balance.
    • Return to the original observation. Explain what the result supports, which conditions it assumes, and one way to test a competing explanation.

    Warn:

At zero net water movement no water molecules cross the membrane. This claim is false: Blotting removes surface solution. Weighing a wet cylinder without blotting adds liquid that did not enter the cells. Repeats reduce random variation but do not correct a miscalibrated balance.

    Key:

    • organelle(細胞小器官):特定の機能を持つ膜に包まれた細胞構造;compartmentalization(区画化):細胞をそれぞれの条件を持つ膜に包まれた領域に分割すること。
Organelles and compartmentalization: original worked-case diagram

Digestion 消化, transport and health evidence

    English

    What would explain this observation?

    • Two foods can have the same mass but provide different nutrients. A health claim must distinguish the nutrient measured from the health outcome inferred.
    • Start with a prediction. State the quantities or features you would compare, then decide what evidence could distinguish two explanations.

    Build the model

    • Large insoluble food molecules are digested into smaller soluble molecules. Carbohydrases form sugars, proteases form amino acids, and lipases form fatty acids and glycerol. Bile emulsifies lipids and helps neutralize acidic stomach contents.
    • digestion: Breakdown of large food molecules; absorption 吸收: Movement of soluble products into the body.
Digestion, transport and health evidence: original worked-case diagram

Choose evidence that can test it

    • Absorption moves soluble products into blood or lymph. Thin exchange surfaces and a large surface area shorten diffusion paths and increase transfer. Enzyme activity and transport are different processes.
    • Use Benedict reagent with controlled heating for reducing sugars, iodine for starch, Biuret reagent for protein, and the ethanol emulsion test for lipids. Keep ethanol away from flames. Use positive and negative controls.

    Work from known quantities

    • State the known values and their units. Choose the relation because its assumptions fit this case, then rearrange before substitution.
    • Known: 6 of 24 study participants report a condition. Proportion = cases / total. Percentage = 6/24 × 100 = 25%. The percentage describes this sample. It does not establish that one food caused the condition; confounders and how the sample was chosen matter.

    Example:

9 of 36 participants report an outcome. Calculate the percentage. Use the same sequence: known quantities → model → relation → substitution → unit and interpretation.

Check the conclusion and its limits

    • Bile is not an enzyme. A positive food test identifies a component under the test conditions; it does not show that a food is healthy or unhealthy in every diet.
    • Return to the original observation. Explain what the result supports, which conditions it assumes, and one way to test a competing explanation.

    Warn:

A correlation between diet and illness proves causation. This claim is false: Bile is not an enzyme. A positive food test identifies a component under the test conditions; it does not show that a food is healthy or unhealthy in every diet.

Supported SL core focus. First assessment 2025; current subject brief acquired; full Biology guide not acquired. Remaining guide, assessment and practical requirements retain their recorded holds.

Prerequisites: transpiration from B.2.3 onwards and natural selection from D.4.1. Each lesson develops its own method before independent transfer.

These are original or explicitly fictional teaching examples, not actual measurements or completed assessed learner investigations.

    14.2

    Adaptation to environment 适应

    English

    What would explain this observation?

    • A cactus survives months without rain while a water lily lives with its leaves flat on the water surface. Both are plants, yet their bodies are nearly opposite. Why?
    • Start with a prediction. State the quantities or features you would compare, then decide what evidence could distinguish two explanations.

    Build the model

    • An adaptation is an inherited feature that improves survival or reproduction in a particular environment. Adaptations are structural (rolled leaves, sunken stomata in a xerophyte 旱生植物), physiological (concentrated urine in a desert mammal) or behavioural (a lizard basking to warm up).
    • A useful adaptation explanation links feature → effect → advantage: rolled leaves trap moist air, so less water is lost by transpiration, so the plant survives drought. Surface area to volume ratio (SA:V) explains many adaptations: a smaller body has a larger SA:V and loses heat or water faster.
    • adaptation: An inherited feature improving survival or reproduction in a given environment; xerophyte: A plant adapted to dry conditions; surface area to volume ratio: Surface area divided by volume, controlling exchange rates.
Adaptation to environment: original worked-case diagram

Choose evidence that can test it

    • Compare water loss from a xerophyte leaf and a mesophyte leaf under the same warm, dry, moving air: the adaptation model predicts less loss from the xerophyte. Keep leaf size, temperature and humidity constant, or the comparison proves nothing.
    • Measure the SA:V of model cubes or of organisms and compare with their habitats; a claim about surface area must come from measured dimensions, not from looking at the picture.

    Work from known quantities

    • State the known values and their units. Choose the relation because its assumptions fit this case, then rearrange before substitution.
    • Known: cube side s = 3 cm. Surface area = 6s² = 6 × 9 = 54 cm²; volume = s³ = 27 cm³. SA:V = 54/27 = 2 per cm. For a 1 cm cube: SA:V = 6/1 = 6 per cm. Tripling the side cut the ratio to one third — larger bodies have less surface per unit volume, so they lose less heat per unit of tissue.

    Example:

A desert mammal model has body surface 400 cm² and volume 2,000 cm³. Find its SA:V and compare it with a small desert mouse at 2 per cm, using the same sequence: known quantities → model → relation → substitution → unit and interpretation. (Answer: SA:V = 400/2,000 = 0.2 per cm — one tenth of the mouse's, so the larger animal loses proportionally less water and heat through its surface.)

Check the conclusion and its limits

    • A plausible story about a feature's use is a hypothesis until evidence supports it; some features are side effects of other traits, not adaptations. The cube model assumes a uniform shape — real organisms extend surface where exchange is needed (lungs, gills, root hairs), breaking the simple scaling.
    • Return to the original observation. Explain what the result supports, which conditions it assumes, and one way to test a competing explanation.

    Warn:

A desert plant adapts during one hot summer by growing thicker cuticle because it needs to. This claim is false: adaptations are INHERITED features shaped by selection over generations; an individual's one-season response is acclimatization, not adaptation, and need never directs the change.

    Key:

Adaptation to environment: inherited features link to survival through a feature → effect → advantage chain, testable by controlled comparison. SA:V = 6/s for a cube, and it falls as size rises — exchange surfaces and body plans follow from it.

Choose evidence that can test it

    • Measure rate using product formed per unit time or a fixed endpoint. For an endpoint test, 1/time is a rate proxy if the same amount of product or substrate change defines the endpoint each time.
    • For starch digestion, equilibrate enzyme and starch in a water bath, control pH with buffer, mix measured volumes, and test samples with iodine at fixed intervals. Use a clean spot for each test. Do not put iodine into the reaction mixture.

    Work from known quantities

    • State the known values and their units. Choose the relation because its assumptions fit this case, then rearrange before substitution.
    • Known: the endpoint is reached in 40 s. Use rate proxy = 1/time. Rate proxy = 1/40 = 0.025 per second. At 20 s the proxy is 0.050 per second, twice as large. This comparison is valid only if the same endpoint and starting concentrations are used.

    Example:

A fixed endpoint takes 50 s. Calculate the rate proxy 1/time. Use the same sequence: known quantities → model → relation → substitution → unit and interpretation.

Check the conclusion and its limits

    • An optimum is specific to the enzyme and conditions. Low temperature usually slows the reaction; it does not necessarily denature the enzyme. Endpoint intervals limit precision.
    • Return to the original observation. Explain what the result supports, which conditions it assumes, and one way to test a competing explanation.

    Warn:

A low temperature always permanently denatures an enzyme. This claim is false: An optimum is specific to the enzyme and conditions. Low temperature usually slows the reaction; it does not necessarily denature the enzyme. Endpoint intervals limit precision.

    Key:

    English

    What would explain this observation?

    • A muscle can use oxygen while you exercise without producing a flame. Respiration transfers energy through a controlled series of reactions.
    • Start with a prediction. State the quantities or features you would compare, then decide what evidence could distinguish two explanations.

    Build the model

    • Aerobic respiration uses oxygen and releases carbon dioxide and water from organic substrates. Energy released can support ATP formation. ATP hydrolysis can be coupled to processes such as active transport and muscle contraction.
    • respiration: Cell reactions that transfer energy from substrates; ATP: A molecule that couples energy transfers in cells.
Respiration, ATP and energy transfers: original worked-case diagram

Choose evidence that can test it

    • Anaerobic processes allow ATP production when oxygen supply cannot support the required aerobic rate, but give less ATP per glucose. In humans lactate can accumulate; yeast can produce ethanol and carbon dioxide.
    • A respirometer can measure oxygen uptake when carbon dioxide is absorbed. Control temperature with a water bath and use a comparison containing inert material. Keep absorbent separated from organisms and follow the school risk assessment.

    Work from known quantities

    • State the known values and their units. Choose the relation because its assumptions fit this case, then rearrange before substitution.
    • Known: oxygen uptake is 0.80 cubic centimetres in 4.0 minutes for 2.0 g of tissue. Rate per mass = volume / (time × mass). Rate = 0.80/(4.0 × 2.0) = 0.10 cubic centimetres per minute per gram. Normalizing allows a fairer comparison of samples of different mass.

    Example:

0.60 cubic centimetres are used in 3.0 minutes by 2.0 g of tissue. Find the mass-specific rate. Use the same sequence: known quantities → model → relation → substitution → unit and interpretation.

Check the conclusion and its limits

    • Breathing ventilates the lungs; respiration consists of chemical reactions in cells. A moving respirometer marker can also reflect temperature or pressure changes.

    Disease transmission and immune response

    English

    What would explain this observation?

    • An antibiotic may help against a bacterial infection but fail against a viral illness. Treatment depends on the causal organism and the evidence supporting diagnosis.
    • Start with a prediction. State the quantities or features you would compare, then decide what evidence could distinguish two explanations.

    Build the model

    • Pathogens cause infectious disease. Physical barriers, phagocytosis and specific immune responses reduce infection. Antibodies bind particular antigens. Vaccination exposes the immune system to antigen 抗原 safely enough to develop memory.
    • pathogen 病原体: An agent that causes disease; antigen: A structure recognized by a specific immune response.
Disease transmission and immune response: original worked-case diagram

Choose evidence that can test it

    • After vaccination, memory cells can support a faster secondary response. Antibiotic resistance arises through heritable variation and selection; an individual bacterium does not choose to become resistant because it needs to survive.
    • Use published infection data to compare rates per equal population size. Distinguish prevalence at a time from new cases over a period. In school, use safe simulations or approved cultures rather than collecting unknown pathogens.

    Work from known quantities

    • State the known values and their units. Choose the relation because its assumptions fit this case, then rearrange before substitution.
    • Known: 18 cases occur in 900 people. Rate per 1,000 = cases / population × 1,000. Rate = 18/900 × 1,000 = 20 cases per 1,000. A second group with 10 cases in 250 people has 40 per 1,000, even though it has fewer cases.

    Example:

12 cases occur among 600 people. Find cases per 1,000 people. Use the same sequence: known quantities → model → relation → substitution → unit and interpretation.

Check the conclusion and its limits

    • Antibiotics do not act on viruses in the same way they act on bacteria. A vaccine is not an immediate cure for an established infection.
    • Return to the original observation. Explain what the result supports, which conditions it assumes, and one way to test a competing explanation.

    Warn:

Every antibiotic is effective against every viral infection. This claim is false: Antibiotics do not act on viruses in the same way they act on bacteria. A vaccine is not an immediate cure for an established infection.

Sampling populations without choosing the answer

    English

    What would explain this observation?

    • A field edge looks richer in plants than its centre. Choosing only the richest patches would build the desired result into the sampling method.
    • Start with a prediction. State the quantities or features you would compare, then decide what evidence could distinguish two explanations.

    Build the model

    • A population 种群 consists of organisms of one species in a defined area. Random quadrats estimate density without deliberately selecting patches. A transect investigates change along an environmental gradient.
    • quadrat 样方: A defined area used for sampling; population: Organisms of one species in a defined area.
Sampling populations without choosing the answer: original worked-case diagram

Choose evidence that can test it

    • Estimate total abundance by multiplying mean density by area, with consistent units. This assumes sampled areas represent the habitat. Patchiness and too few samples widen uncertainty.
    • Choose coordinates with random numbers before visiting the patches. Record quadrat area and counting rules. For a transect, use fixed distances and measure a relevant abiotic variable. Do not damage habitats or sample unsafe locations.

    Work from known quantities

    • State the known values and their units. Choose the relation because its assumptions fit this case, then rearrange before substitution.
    • Known: five 0.25 square metre quadrats contain 3, 4, 6, 5 and 2 plants. Mean count = 20/5 = 4. Density = 4/0.25 = 16 plants per square metre. Estimated abundance in 100 square metres = 16 × 100 = 1,600 plants.

    Example:

Mean count is 6 in a 0.5 square metre quadrat. Find density per square metre. Use the same sequence: known quantities → model → relation → substitution → unit and interpretation.

Check the conclusion and its limits

    • Quadrats suit organisms that do not move quickly. A food chain arrow shows the direction of energy transfer, not the direction a predator travels.
    • Return to the original observation. Explain what the result supports, which conditions it assumes, and one way to test a competing explanation.

    Warn:

Sampling only the most crowded patches gives an unbiased population estimate. This claim is false: Quadrats suit organisms that do not move quickly. A food chain arrow shows the direction of energy transfer, not the direction a predator travels.

    Protein synthesis

    English

    What would explain this observation?

    • A gene hundreds of bases long produces a much shorter protein, and a one-base mutation can destroy the whole product. How does the cell read a base sequence into an amino-acid sequence?
    • Start with a prediction. State the quantities or features you would compare, then decide what evidence could distinguish two explanations.

    Build the model

    • In transcription 转录, the enzyme RNA polymerase builds an mRNA strand complementary to one DNA template strand. In translation 翻译, a ribosome reads the mRNA in groups of three bases — codons 密码子 — and joins the matching amino acids into a polypeptide.
    • Each codon specifies one amino acid through the genetic code, which is degenerate: several codons can give the same amino acid. A stop codon 终止密码子 ends the chain and codes for nothing. Because the reading frame runs in threes, inserting or deleting one base shifts every later codon — a frameshift.
    • transcription: Copying a gene's DNA sequence into a complementary mRNA; translation: Reading mRNA codons at a ribosome to build a polypeptide; codon: A group of three mRNA bases specifying one amino acid; stop codon: A codon that ends the chain without coding for an amino acid.
    Protein synthesis: original worked-case diagram

Choose evidence that can test it

    • The base sequence of a gene predicts the amino-acid sequence of its protein through the code table; compare a known gene and its protein to check. Radioactive-labelling experiments placed amino acids in the order the mRNA specified — evidence for the codon reading, not a guess.
    • Mutations that change a codon to a stop codon truncate the protein exactly at that point, which can be checked against the sequence.

    Work from known quantities

    • State the known values and their units. Choose the relation because its assumptions fit this case, then rearrange before substitution.
    • Known: a coding mRNA segment has 63 bases, including one stop codon. Codons = bases / 3 = 63 / 3 = 21. The stop codon codes for no amino acid, so the peptide has 21 − 1 = 20 amino acids.

    Example:

A coding segment of 90 bases includes one stop codon. Find the peptide length using the same sequence: known quantities → model → relation → substitution → unit and interpretation. (Answer: codons = 90/3 = 30; peptide = 30 − 1 = 29 amino acids. Bases are not amino acids — divide by three first.)

Check the conclusion and its limits

    • This counting model covers the coding sequence only: real genes also carry promoters, untranslated regions and, in eukaryotes, introns that are removed before translation. A gene's total base count is therefore not a peptide-length calculation.
    • Return to the original observation. Explain what the result supports, which conditions it assumes, and one way to test a competing explanation.

    Warn:

Each base in the coding sequence codes for one amino acid. This claim is false: amino acids are coded by triplets — three bases per codon — and the stop codon adds none, so a 63-base segment gives 20 amino acids, not 63.

    Key:

Protein synthesis: transcription copies the gene into mRNA; translation reads codons in threes at the ribosome. Codons = bases ÷ 3, and the stop codon codes for no amino acid; a one-base insertion or deletion shifts the whole reading frame.

    Scope and prerequisites

    English

Prerequisites: DNA structure from A.1.2 and protein synthesis from D.1.2. Each lesson develops its own method before independent transfer.

These are original or explicitly fictional teaching examples, not actual measurements or completed assessed learner investigations.

    26.2

    Mutations and gene editing 突变

    English

    What would explain this observation?

    • One changed base in the haemoglobin gene causes sickle cell disease, while a one-base deletion in another gene destroys the entire protein. Why does one small edit matter so much more than the other?
    • Start with a prediction. State the quantities or features you would compare, then decide what evidence could distinguish two explanations.

    Build the model

    • A mutation is a change in the base sequence of DNA. A substitution 替换 swaps one base; if the new codon still codes for the same amino acid (a silent change) the protein is unchanged, but a change to a different amino acid can alter the protein's shape, as in sickle cell disease.
    • An insertion or deletion shifts the reading frame 阅读框: every codon after the change is misread, so the protein is usually destroyed. Gene editing 基因编辑 with CRISPR-Cas9 uses a guide RNA to find one chosen sequence and the Cas9 enzyme to cut both strands there; the cell's repair then disables or rewrites the gene.
    • mutation: A change in the base sequence of DNA; substitution: A mutation swapping one base for another; reading frame: The grouping of bases into codons from a fixed start; gene editing: Deliberately changing a chosen DNA sequence, for example with CRISPR-Cas9.
Mutations and gene editing: original worked-case diagram

Choose evidence that can test it

    • Compare the DNA sequence with the protein produced: a substitution predicts one changed amino acid at one position, while a deletion predicts every amino acid changed after it. Sequence data decide between the two.
    • In gene editing, sequencing the target site after repair shows whether the cut disabled the gene or inserted the intended change; an off-target check sequences similar sites elsewhere.

    Work from known quantities

    • State the known values and their units. Choose the relation because its assumptions fit this case, then rearrange before substitution.
    • Known: the coding sequence AUG CCU GGA UAA reads in codons of three. Delete the fourth base (the first C). The sequence becomes AUG CCG GAU AA: the first codon is intact, but every later codon is shifted, and the stop codon is destroyed — the ribosome reads on into wrong codons. A substitution of that same C to A instead gives AUG CAU GGA UAA: only the second codon changes.

    Example:

A coding sequence of 30 bases loses one base near the start. How many codons are potentially misread, using the same sequence: known quantities → model → relation → substitution → unit and interpretation? (Answer: 30/3 = 10 codons; losing one base near the start shifts all 10, while a substitution would have changed at most one.)

Check the conclusion and its limits

    • This simple model covers the coding region; mutations in control regions can change how MUCH protein is made without changing its sequence. Gene editing's safety depends on off-target rates measured experimentally, not on the guide's design alone.
    • Return to the original observation. Explain what the result supports, which conditions it assumes, and one way to test a competing explanation.

    Warn:

A mutation is always harmful and always changes the protein. This claim is false: silent substitutions change the DNA but not the amino acid, many mutations fall outside coding regions, and a few mutations are beneficial — variation from mutation is the raw material of evolution.

    Key:

Mutations and gene editing: substitutions change one codon; insertions and deletions shift the reading frame and usually destroy the protein. CRISPR-Cas9 edits by guide-RNA targeting plus a two-strand cut, with results checked by sequencing.

    Homeostasis 稳态

    English

    What would explain this observation?

    • After a large meal your blood glucose jumps, yet within an hour it returns to near where it started — and after a night without food it still does not collapse. What keeps it steady?
    • Start with a prediction. State the quantities or features you would compare, then decide what evidence could distinguish two explanations.

    Build the model

    • Homeostasis is the maintenance of a nearly constant internal environment around a set point. It runs on negative feedback 负反馈: a detector notices the change, a control centre compares it with the set point, and an effector produces a response that OPPOSES the change.
    • For blood glucose: when it rises, the pancreas releases insulin 胰岛素, which makes liver and muscle cells take up glucose and store it as glycogen, so glucose falls. When glucose falls too low, glucagon 胰高血糖素 triggers the liver to convert glycogen back to glucose, so it rises. Each hormone switches off as the level returns to the set point.
    • homeostasis: Maintaining a nearly constant internal environment around a set point; negative feedback: A control loop whose response opposes the original change; insulin: The hormone that lowers blood glucose by promoting uptake and storage; glucagon: The hormone that raises blood glucose by releasing stored glucose.
    Homeostasis: original worked-case diagram

Choose evidence that can test it

    • Measure blood glucose at intervals after a meal: the model predicts a rise, a peak, and a return towards the set point as insulin acts — and the same record shows insulin rising before glucose falls, the cause arriving before the correction.
    • Compare a person whose pancreas cannot make insulin: glucose stays high after a meal, direct evidence that insulin is the link between the detector and the response.

    Work from known quantities

    • State the known values and their units. Choose the relation because its assumptions fit this case, then rearrange before substitution.
    • Known: a person's blood glucose rises from a set point of 5.0 to 7.0 mmol/L after a meal. Change = final − initial = 7.0 − 5.0 = 2.0 mmol/L. Percentage deviation = 2.0/5.0 × 100 = 40% above the set point. Negative feedback acts until the value returns close to 5.0 mmol/L.

    Example:

During a fast, blood glucose falls from 5.0 to 4.2 mmol/L. Find the percentage deviation from the set point and name the responding hormone, using the same sequence: known quantities → model → relation → substitution → unit and interpretation. (Answer: change = 4.2 − 5.0 = −0.8 mmol/L; deviation = −0.8/5.0 × 100 = −16%; glucagon responds to raise it.)

Check the conclusion and its limits

    • Homeostasis is NEAR-constant, not fixed: the value oscillates around the set point because each response takes time. This model covers blood glucose; temperature, water and carbon dioxide each have their own detectors and effectors with the same logic.
    • Return to the original observation. Explain what the result supports, which conditions it assumes, and one way to test a competing explanation.

    Warn:

Negative feedback makes the measured value negative. This claim is false: "negative" means the response OPPOSES the change — a rise triggers a fall and a fall triggers a rise. The measured value stays positive and close to the set point.

    Key:

Homeostasis: detector → control centre → effector loops keep internal conditions near a set point by negative feedback. Insulin lowers raised blood glucose; glucagon raises lowered blood glucose; the loop switches off as balance returns.

    32.1

    Scope and prerequisites

    English

Supported SL core focus. First assessment 2025; current subject brief acquired; full Biology guide not acquired. Remaining guide, assessment and practical requirements retain their recorded holds.

Prerequisites: variation and inheritance from D.1 and evolution evidence from A.4.1. Each lesson develops its own method before independent transfer.

These are original or explicitly fictional teaching examples, not actual measurements or completed assessed learner investigations.

    32.2

    Natural selection

    English

    What would explain this observation?

    • After several dry summers, most plants in a population have deeper roots than their grandparents' generation did. Nobody chose this — how did the population change?
    • Start with a prediction. State the quantities or features you would compare, then decide what evidence could distinguish two explanations.

    Build the model

    • Natural selection needs three ingredients: inherited variation 变异 exists in the population; a selection pressure 选择压力 makes some variants survive and reproduce better than others; and the successful variants' alleles become commoner in the next generation. Over generations the population's traits shift.
    • Selection does not create mutations and no individual evolves: random mutation supplies the variation BEFORE the pressure acts, and it is the population's allele frequency 等位基因频率 that changes, not any one organism.
    • natural selection: The process by which heritable variants that fit the environment leave more offspring, changing allele frequencies over generations; variation: Inherited differences among individuals of a population; selection pressure: An environmental factor that affects survival or reproduction; allele frequency: The proportion of one allele among all copies of that gene in a population.
    Natural selection: original worked-case diagram

Choose evidence that can test it

    • The model predicts the direction of change: after repeated droughts, measure the deep-root allele's frequency in stored seed collections or dated samples — it should rise. If it does not, the explanation fails.
    • A fair test compares the SAME population across time or matched populations across a boundary, so that migration or sampling differences cannot fake the shift.

    Work from known quantities

    • State the known values and their units. Choose the relation because its assumptions fit this case, then rearrange before substitution.
    • Known: before the droughts, 200 gene copies include 90 copies of the deep-root allele; after several generations, 120 of 200. Allele frequency = copies of the allele / total copies. Before: 90/200 = 45%. After: 120/200 = 60%. The frequency rose by 15 percentage points — evidence of selection acting on the population.

    Example:

In a new generation, 140 of 200 copies carry the deep-root allele. Find the frequency and the change from the previous generation using the same sequence: known quantities → model → relation → substitution → unit and interpretation. (Answer: 140/200 = 70%; change = 70% − 60% = +10 percentage points. Frequencies of two alleles at one gene sum to 100%.)

Check the conclusion and its limits

    • A frequency shift shows association between the trait and survival; proving the mechanism needs the variation to be heritable and the survival difference measured. Genetic drift in small populations can shift frequencies with no selection at all.
    • Return to the original observation. Explain what the result supports, which conditions it assumes, and one way to test a competing explanation.

    Warn:

An organism develops a useful adaptation during its lifetime because it needs one. This claim is false: mutation is random with respect to need — the deep-root alleles existed before the drought. Selection only changes which variants leave more offspring; individuals do not evolve.

    Key:

Natural selection: inherited variation + a selection pressure + differential reproduction shifts allele frequencies over generations. Track the shift with allele counts divided by total copies, and remember that the population evolves — never the individual.

    33.1

    Scope and prerequisites

    English

Supported SL core focus. First assessment 2025; current subject brief acquired; full Biology guide not acquired. Remaining guide, assessment and practical requirements retain their recorded holds.

Prerequisites: populations and ecosystems from D.4.1 and energy flow from earlier ecology. Each lesson develops its own method before independent transfer.

These are original or explicitly fictional teaching examples, not actual measurements or completed assessed learner investigations.

    33.2

    Sustainability and change

    English

    What would explain this observation?

    • A fishery that harvests the same tonnage every year can thrive for decades, while a neighbouring one collapses in five. Both fish the same sea — what separates them?
    • Start with a prediction. State the quantities or features you would compare, then decide what evidence could distinguish two explanations.

    Build the model

    • Populations cannot grow without limit. The logistic model 逻辑斯蒂模型 shows growth slowing as resources run short, levelling at the environment's carrying capacity 环境容纳量 — the largest population the habitat can support. Food, space, disease and predation supply the limits.
    • A renewable resource 可再生资源 is sustainable only when harvest stays at or below regrowth. Change is constant in ecosystems: climate shifts, species arrive, and human activity alters habitats, so management must track stocks and adjust.
    • logistic model: Growth that levels at the carrying capacity as resources become limiting; carrying capacity: The largest population an environment can support; renewable resource: A resource that regenerates, such as a fish stock or a forest.
Sustainability and change: original worked-case diagram

Choose evidence that can test it

    • Compare the stock before and after each harvest season: a sustainable fishery holds the stock near its level, while decline year on year means harvest exceeds regrowth. Distinguish a falling stock from a falling CATCH, which can hide the decline until late.
    • Track the same population over several years rather than once; a single count cannot show a trend.

    Work from known quantities

    • State the known values and their units. Choose the relation because its assumptions fit this case, then rearrange before substitution.
    • Known: a fish stock is 1,000 tonnes and regrows 100 tonnes per year. A harvest of 150 tonnes per year gives net change = regrowth − harvest = 100 − 150 = −50 tonnes per year. After two years the stock is 1,000 − 2 × 50 = 900 tonnes — a sustainable harvest at this regrowth is at most 100 tonnes per year.

    Example:

After better management the regrowth is 120 tonnes per year and the harvest quota is set at 80 tonnes per year. Find the yearly change in the stock and the stock after three years from 900 tonnes, using the same sequence: known quantities → model → relation → substitution → unit and interpretation. (Answer: net change = 120 − 80 = +40 tonnes per year; after three years, 900 + 3 × 40 = 1,020 tonnes — the stock recovers.)

Check the conclusion and its limits

    • The balance model assumes a constant regrowth rate; in reality regrowth depends on the stock size itself, collapsing fastest when the stock is already small. One year's figures cannot establish a trend — management needs the whole time series and honest catch reporting.
    • Return to the original observation. Explain what the result supports, which conditions it assumes, and one way to test a competing explanation.

    Warn:

A renewable resource cannot be used up. This claim is false: renewable means it CAN regenerate, not that it always outpaces use — a stock harvested faster than its regrowth shrinks every year and can collapse entirely, as overfished cod stocks did.

    Key:

Sustainability and change: populations level at the carrying capacity, and renewable resources stay only if harvest ≤ regrowth. Track stocks over time and set harvests from measured regrowth, because ecosystems never stand still.
